Senna (Tinnevelly senna fruits), a known laxative derived from plants, was administered by gavage to Sprague-Dawley (Crl:CD (SD) BR) rats once daily at dose levels of 0, 25, 100 and 300 mg/kg/day for up to 104 consecutive weeks. Based upon clinical signs related to the laxation effect of senna, the highest dose (300 mg/kg/day) was considered to be a maximum tolerated dose. Sixty animals per sex were assigned to the control and dose groups. Assessments included clinical chemistry, hematology, full histology (control and high-dose groups; in addition, low and mid dose: intestinal tract, adrenals, liver, kidneys, brain and gross lesions) and toxicokinetics. Endothelin-1 (ET-1), a potent endothelium-derived vasoconstrictor peptide, is secreted in response to insulin. Elevated circulating ET-1 levels have been found in patients with diabetes mellitus and vascular dysfunction. The question arises whether ET-1 acts as a direct modulator of insulin secretion. To test this, we studied the effects of ET-1 on isolated mouse islets of Langerhans. ET-1 (1 nmol/l-1 Μmol/l) dose-dependently stimulated insulin secretion from islets incubated in the presence of 16.7 mmol/l glucose (p<0.05). The effect of ET-1 is glucose-dependent since no potentiation was found at 3.3 mmol/l glucose. Furthermore, ET-1 induced a large, transient increase in glucose-stimulated insulin secretion during islet perifusion in the presence (p<0.001), but not in the absence, of extracellular Ca2+. The rate of 45Ca2+-efflux from 45Ca2+-prelabelled islets was transiently stimulated by ET-1 during perifusion at 16.7 mmol/l glucose in the presence of extracellular Ca2+ (p<0.001). A short-lived increase in 45Ca2+-efflux was also observed in the absence of extracellular Ca2+ (p<0.05). It is suggested that the effects of ET-1 on insulin secretion are critically dependent on influx via Ca2+-channels. In addition, ET-1 transiently enhanced 86Rb+-efflux from 86Rb+-prelabelled islets both in the presence (p<0.001) and in the absence (p<0.001) of extracellular Ca2+ suggesting that ET-1 does not elicit insulin secretion by inhibition of the potassium permeability. Our study provides evidence that ET-1 stimulates insulin secretion via a direct effect on the islets of Langerhans.

Hyperkalemia is a common electrolyte abnormality that may be difficult to manage because of a lack of effective therapies. Sodium zirconium cyclosilicate is a nonabsorbed cation exchanger that selectively binds potassium in the intestine. To evaluate the efficacy and safety of zirconium cyclosilicate for 28 days in patients with hyperkalemia. An imbalance in electrolyte concentration during separation from cardiopulmonary bypass (CPB) may lead to a disruption in excitation-contraction coupling resulting in a failure to wean. The etiology of myocardial dysfunction is multifactorial, and includes alterations in acid-base balance, glucose metabolism, and cellular function. The purpose of this study was to assess the effect of hyperkalemia on myocardial function during separation from CPB. A porcine model (n = 5) of hypothermic (32 degrees C) CPB was used where hyperkalemia [K+ (6.5 +/- 1.0)] was created before weaning. A 3-minute weaning process was initiated once normothermia was achieved. Mixed venous and arterial samples were obtained during CPB, weaning, and 10 minutes postbypass. An efficient and direct procedure has been developed for the preparation of amidoalkyl naphthols by a one-pot condensation of aryl aldehydes, β-naphthol and urea or amides, in the presence of potassium dodecatungstocobaltate trihydrate [K5CoW12O40 · 3H2O, (1 mol%)] as a heterogeneous catalyst. The reactions were carried out in 1,2-dichloroethane at room temperature or under solvent-free media at elevated temperature. The present methodology offers several advantages such as excellent yields, simple procedure and the catalyst exhibited remarkable reusability.

This study evaluated the potential of glauconitic sandstone as a fertilizer for supplying potassium to plants. The glauconite sandstone (Maraveh, Iran), as analyzed by X-ray fluorescence, was composed of 2.24% potassium oxide plus high contents of silicon, aluminum and ferric oxide. One-year old olive trees, Olea europaea L., were grown in sand or hydroponic culture in a greenhouse under three potassium treatments. Modified Hoagland nutrient solutions based on potassium treatments including 0.5 mM K+, 5 mM K+ and 400 g glauconitic sandstone powders (per 10 L in hydroponics and per 2.5 L in sand instead of K+ supply) were used in both cultures. Plants grown under the three different potassium treatments did not show any potassium deficiency symptoms. In the sand culture, growth and potassium content were higher in plants fed with 5 mM potassium than with the other two potassium treatments. Growth retardation and decreased potassium content in plants fed with 0.5 mM potassium were more severe in the hydroponic culture than in the sand culture. However, plants fed with 400 g glauconitic sandstone showed higher growth in the hydroponic culture than the sand culture. Thus, glauconitic sandstone has the ability to release potassium and can be utilized in combination with other potassium fertilizers.

An investigation of the electrochemical activity of human white blood cells (WBC) for biofuel cell (BFC) applications is described. WBCs isolated from whole human blood were suspended in PBS and introduced into the anode compartment of a proton exchange membrane (PEM) fuel cell. The cathode compartment contained a 50 mM potassium ferricyanide solution. Average current densities between 0.9 and 1.6 μA cm-2 and open circuit potentials (Voc) between 83 and 102 mV were obtained, which were both higher than control values. Cyclic voltammetry was used to investigate the electrochemical activity of the activated WBCs in an attempt to elucidate the mechanism of electron transfer between the cells and electrode. The mTOR pathway orchestrates cellular homeostasis. The rapamycin-sensitive mTOR complex (mTORC1) in the kidney has been widely studied; however, mTORC2 function in renal tubules is poorly characterized. Here, we generated mice lacking mTORC2 in the distal tubule (Rictorfl/fl Ksp-Cre mice), which were viable and had no obvious phenotype, except for a 2.5-fold increase in plasma aldosterone. Challenged with a low-Na+ diet, these mice adequately reduced Na+ excretion; however, Rictorfl/fl Ksp-Cre mice rapidly developed hyperkalemia on a high-K+ diet, despite a 10-fold increase in serum aldosterone levels, implying that mTORC2 regulates kaliuresis. Purpose This study compares the in vivo relative lung bioavailability of Hydrofluoroalkane (HFA) Seretide delivered via unprimed and unwashed Aerochamber Plus (AP) or Volumatic (VM) spacers, a integrated breath-actuated vortex Synchro-Breathe (SB) device and an Evohaler pMDI (EH) device using adrenal suppression and early fall in serum potassium (K) as surrogates for respirable dose. Methods Seventeen healthy volunteers completed this randomised double-blind, double-dummy crossover study. Single doses of placebo/Seretide 250 (total dose ex valve fluticasone 2000 mcg/salmeterol 200 mcg) were administered via the devices. Overnight urinary cortisol/creatinine (OUCC) and serum K were measured at baseline and after each dose. Results Significant suppression of OUCC and K occurred from baseline with the SB, AP and VM but not with the EH devices. The geometric mean fold suppression (95% confidence interval, p) was: EH, 1.59 (0.80–3.14, p = 0.40); AP, 4.26 (3.01–6.02, p < 0.001); VM, 3.11 (1.99–4.78, p < 0. 001); SB, 3.29 (2.04–5.24, p < 0.001). For K, the arithmetic mean fall (mmol/l) (95% confidence interval; p) was: EH, −0.10 (−0.25–0.05, p = 0.18); AP, −0.23 (−0.41 to −0.04, p = 0.02); VM, −0.22 (−0.44 to −0.01, p = 0.04); SB, −0.28 (−0.42 to −0.13, p = 0.001). Conclusions The breath-actuated SB device was comparable to ‘out of the box’ small and large volume spacers and produced similar improvements in relative systemic lung bioavailability for fluticasone and salmeterol.

Status of organic carbon (C), total nitrogen (N), available potassium (K), calcium (Ca) and phosphorus (P) in three different depths (0−5 cm, 5−15 cm and 15−30 cm) on two hill slopes of 35% and 55% in orange orchard cultivated by the Mro tribe of Chittagong Hill Tracts (CHTs) were evaluated and compared with those in degraded bush forests, through digging three profiles in each land use. The content of all the five nutrients was found to be higher in the soil of orange orchard than in the soil of forest. But the variation was not consistent for both the slopes. The content varied depth wise also, having the highest value in surface soil in case of both the land uses on both the slopes. A mean available K content was significantly higher in orange orchard than in forest on 55% slope, while it was lower on 35% slope.
