    Significance In diving birds like penguins, physiologic considerations suggest that increased hemoglobin (Hb)-O 2 affinity may improve pulmonary O 2 extraction and enhance dive capacity. We integrated experimental tests on whole-blood and native Hbs of penguins with protein engineering experiments on reconstructed ancestral Hbs. The experiments involving ancestral protein resurrection enabled us to test for evolved changes in Hb function in the stem lineage of penguins after divergence from their closest nondiving relatives. We demonstrate that penguins evolved an increased Hb-O 2 affinity in conjunction with a greatly augmented Bohr effect (i.e., reduction in Hb-O 2 affinity at low pH) that should maximize pulmonary O 2 extraction without compromising O 2 delivery at systemic capillaries.

    Persistent organic pollutants (POPs) pose a health risk for southern resident killer whales (SRKWs). Data on maternal contaminant transfer to calves are needed to inform models that estimate future contaminant loads as well as assess risk to newborn killer whale calves exposed to POPs. We conducted a study on trained female killer whales and their calves to fill these data gaps. POPs and lipid content were quantified in blood serum (POP levels in serum and blubber are highly correlated) collected during gestation and in milk and serum collected post-partum from female killer whales. Serum samples were also collected from calves during the lactation period. POP concentrations in milk collected over 15 months post-partum from the primiparous female decreased by 47-65%, depending on the contaminant class. The highest influx of contaminants to calves tended to occur soon after birth. Greater contaminant transfer rates during early lactation were also reflected in maternal serum POP levels. POP levels in maternal serum decreased significantly during the first 144-158 days post-partum, depending on contaminant class, and then leveled off through the remaining lactation period. This resulted in 67-81% reductions in POP levels in maternal serum over 15 months. By 15 months post-partum, serum POP levels from the primiparous female had dropped to levels measured in the multiparous female during pregnancy. Dissimilar to the multiparous female, body mass and blubber thickness in the primiparous female also declined significantly during the first 3-5 months post-partum, demonstrating linkages between lipid and POP transfer from blubber stores to milk in early lactation. By the end of lactation, lipid-corrected POP concentrations in serum from the first-born calf were 5-8 times greater than the corresponding POP levels from her primiparous mother. These results demonstrate that very young neonatal SRKW calves, particularly first-born calves, are at high risk from contaminant exposure

    OBJECTIVE To compare serum cardiac troponin I (cTnI) concentrations between sea otters with and without cardiomyopathy and describe 2 cases of cardiomyopathy with different etiologies. ANIMALS 25 free-ranging southern sea otters (Enhydra lutris nereis) with (n = 14; cases) and without (11; controls) cardiomyopathy and 17 healthy managed southern sea otters from aquariums or rehabilitation centers (controls). PROCEDURES Serum cTnI concentration was measured in live sea otters. Histopathologic and gross necropsy findings were used to classify cardiomyopathy status in free-ranging otters; physical examination and echocardiography were used to assess health status of managed otters. Here, we report a multi-faceted investigation of DNA methylation across 11,117 samples from 176 different species. We performed an unbiased clustering of individual cytosines into 55 modules and identified 31 modules related to primary traits including age, species lifespan, sex, adult species weight, tissue type and phylogenetic order. Analysis of the correlation between DNA methylation and species allowed us to construct phyloepigenetic trees for different tissues that parallel the phylogenetic tree. In addition, while some stable cytosines reflect phylogenetic signatures, others relate to age and lifespan, and in many cases responding to anti-aging interventions in mice such as caloric restriction and ablation of growth hormone receptors.     Background Pacific walruses are found in Arctic regions of the Chukchi and Bering Sea where rapid changes in environmental conditions resulting in loss of sea ice are occurring. Therefore, accurate life history data are crucial for species management plans and longitudinal data collected over the lives of individual walruses housed in zoos and aquaria provide otherwise difficult to obtain biological information. Results While similar at birth, Gompertz regression curves indicated that males grew faster than females (p < 0.0001) in weight (99 kg vs 57.6 kg/y) and length (26.9 cm vs 26.3 cm/y) with physical differences being detected by age 3 for weight and age 7 for length.     This investigation compared the fatty acid (FA) levels found in erythrocyte (RBC) membranes, plasma, whole blood (WB), and blubber from wild Alaskan (Bristol Bay) belugas (Delphinapterus leucas) (BBB, n = 9) with oceanaria-based belugas (OBB, n = 14) fed a controlled diet consisting of primarily herring (Clupea harengus) and capelin (Mallotus villosus). FA patterns in RBCs, WB, and plasma varied considerably between BBB and OBB animals. Focusing on RBC FA levels of known dietary origin, the OBBs had markedly higher levels of 20:1n9,11 and 22:1n9,11. RBC levels of these fatty acids were 1% and 0.2% in the BBBs, but 8.2% and 4.5%, respectively, in the OBBs (p < 0.05 both). These long-chain mono-unsaturated FAs (LC-MUFAs) are rich in herring and capelin but not in the prey species (i.e., salmonids, smelt, cod, and shrimp) generally available to BBBs.     Abstract: The pharmacokinetics of danofloxacin was investigated in rehabilitated California brown pelicans (Pelecanus occidentalis californicus) after a single intramuscular injection at a dose of 15 mg/kg body weight. The concentration of the drug in plasma was assayed by high-pressure liquid chromatography. A sparse-sampling design was used to reduce the number of samples (1–4 venipunctures) obtained from 24 brown pelicans. A population pharmacokinetic analysis with nonlinear mixed-effects modeling was used to accommodate the sparse-sampling strategy. The nonlinear mixed-effects modeling approach measured both fixed effects (typical values for the population) and random effects (between-subject variability) for this population. A 1-compartment model best represented the concentration-versus-time data after injection. After injection, the elimination half-life, peak concentration, area under the curve, and volume of distribution were 2.76 hours, 2.5 µg/mL, 13.75 µg/h/mL, and 4.35 L/kg, respectively. Rate of absorption was highly variable among the birds. The intramuscular injection of danofloxacin in pelicans at this dose produced plasma concentrations that meet therapeutic targets for bacteria with a minimum inhibitory concentration of ≤0.25 µg/mL. This dose can be used for future studies to evaluate the efficacy of danofloxacin for treating susceptible bacteria.

    For many reptile species, adequate ultraviolet B (UVB) radiation is essential for proper calcium metabolism. In this study, the effects of UVB radiation on calcium metabolism were evaluated in green sea turtles (Chelonia mydas). 25-Hydroxyvitamin D, parathyroid hormone (PTH), and ionized calcium (iCa) were measured in juvenile (n = 18, 9 indoor, 9 outdoor) and adult (n = 8, 4 indoor, 4 outdoor) turtles. All animals were fed an identical diet. Outdoor animals had access to unfiltered sunlight, whereas indoor animals were housed under artificial lighting without UVB. Mean values for 25-hydroxyvitamin D for the outdoor and indoor groups were 34.33 ± 7.98 nmol/L and 7.11 ± 1.69 nmol/L for juveniles and 73.25 ± 30.34 nmol/L and 14.0 ± 11.52 nmol/L for adults respectively. Mean values for iCa for the outdoor and indoor groups were 0.98 ± 0.07 mmol/L and 0.99 ± 0.06 mmol/L for juveniles and 1.18 ± 0.22 mmol/L and 0.97 ± 0.18 mmol/L for adults respectively. UVB exposure (P < 0.001) and age (P < 0.001) had a significant effect on 25-hydroxyvitamin D as well as a significant interaction between the two variables (P = 0.008), with highest values in adult outdoor turtles. There was a significant interaction between age group and UVB status for iCa (P = 0.036), with greater values in older outdoor turtles. 25-Hydroxyvitamin D and total calcium were positively correlated, rs = 0.39, P = 0.042. iCa and calcium-to-phosphorus ratios were also positively correlated, rs = 0.42, P = 0.027. These results suggest that UVB exposure is an important source of 25-hydroxyvitamin D for green sea turtles and has significant effects on calcium metabolism in this species. PTH values in this study were near the minimum limits of detection and suggest that current mammalian-based PTH assays are not valid for reptiles.

Seal parapoxvirus is currently classified as a tentative species of the Parapoxvirus genus. Tissue or swab samples were analyzed from 11 pinnipeds of different host species undergoing rehabilitation on the east and west coasts of the United States of America (USA) that were positive for parapoxvirus.     Campylobacter pinnipediorum was described recently for isolates recovered from pinnipeds. The novel species was further split into 2 subspecies based on host and geography, with C. pinnipediorum subsp. pinnipediorum recovered from otariid seals in California (USA) and C. pinnipediorum subsp. caledonicus recovered from phocid seals in Scotland. We report details of the infections of 7 pinnipeds from which C. pinnipediorum was isolated: C. pinnipediorum subsp. caledonicus was isolated from 2 harbour seals Phoca vitulina and a single grey seal Halichoerus grypus, and C. pinnipediorum subsp. pinnipediorum was isolated from California sea lions Zalophus californianus. Six of the isolates were recovered from samples collected at post-mortem investigation. In 2 of the Scottish seals and in 3 of the California seals, C. pinnipediorum was the sole bacterial isolate recovered from abscesses present and suggests they may have resulted from conspecific or intraspecific bite wounds.

    ABSTRACTAccurate identification of individual ages within wild bottlenose dolphins (Tursiops truncatus) is critical for determining population health and the development of population management strategies. As such, we analyzed DNA methylation patterns by applying a custom methylation array (HorvathMammalMethyl40) to both blood (n = 140) and skin samples (n = 87) from known age or approximate age (0 to 57 years) bottlenose dolphins. We present three bottlenose dolphin specific age estimation clocks using combined blood and skin (48 CpGs, R = 0.93, median absolute error = 2.13 years), blood only (64 CpGs, R = 0.97, error= 1.46 years) and skin only (39 CpGs, R = 0.95, error= 2.53). Our sex estimator based on 71 CpGs predicts the sex of any odontocete species with 99.5% accuracy. We characterize individual cytosines that correlate with sex and age in dolphins.The presented epigenetic clocks are expected to be useful for conservation efforts and for studying anthropogenic events.

    ABSTRACTAging is often perceived as a degenerative process resulting from random accrual of cellular damage over time. Despite this, age can be accurately estimated by epigenetic clocks based on DNA methylation profiles from almost any tissue of the body. Since such pan-tissue epigenetic clocks have been successfully developed for several different species, we hypothesized that one can build pan-mammalian clocks that measure age in all mammalian species. To address this, we generated data using 11,754 methylation arrays, each profiling up to 36 thousand cytosines in highly-conserved stretches of DNA, from 59 tissue-types derived from 185 mammalian species. From these methylation profiles, we constructed three age predictors, each with a single mathematical formula, termed universal pan-mammalian clocks that are accurate in estimating the age (r>0.96) of any mammalian tissue.     Abstract: Neutrophils are one of the initial cell lines of protection against pathogens, and when their concentrations in the blood are low, animals are highly susceptible to infections. Neutropenia has been reported in cetaceans secondary to administration of systemic sulfa antibiotics or antifungal medications and severe, overwhelming infection. Filgrastim was administered to treat neutropenia over a 15-y period in 11 cetaceans comprising four species—beluga (Delphinapterus leucas, n = 1), bottlenose dolphin (Tursiops truncatus, n = 4), killer whale (Orcinus orca, n = 5), and short-finned pilot whale (Globicephala macrorhynchus, n = 1)] ranging in age from 1 wk to >24 y. Seven study animals received multiple doses (2–6). All animals responded to at least one dose (1–7 µg/kg) of parenteral filgrastim characterized by an increase in peripheral immature (band) neutrophils, segmented neutrophils, or both. In most cases (9/11), neutrophil counts increased within 48 h of a single dose. Duration of response varied but was at least 2 wk in eight of the 11 animals and 5–9 d in the remaining animals. No adverse reactions were observed in any cases.

    Urban coastal ecosystems are unique intersections of human development and biodiversity, and monitoring populations in these areas is critical to understanding ecosystem health and function. Three highly urbanized estuary systems, San Diego Bay, San Gabriel River, and Seal Beach National Wildlife Refuge (all in California), are the northernmost foraging habitats for green turtles ( Chelonia mydas) in the Eastern Pacific. Here, we report blood biochemistry and morphological parameters for these Southern California green sea turtle foraging aggregations to investigate the current health status of these animals and provide baseline values for future work. Morphometric and blood biochemistry parameters for green turtles captured in this study ( n = 39) were clinically reasonable and were generally consistent with previously reported parameters for green turtles.

    Aspergillus is a significant pathogen in zoological species, although information on environmental variables influencing fungal prevalence in zoological settings are lacking. The objective of the study was to estimate the prevalence of and to identify environmental factors associated with aerosolised Aspergillus spores at a zoological park to advance the understanding of fungal exposure as a first step towards improved mitigation strategies for susceptible animals. Twenty-one locations were sampled for presence of Aspergillus species using the SAS Super 180 Microbial Air Sampler, while twenty-two environmental factors were evaluated every two weeks at SeaWorld of California during two 12-month periods.     A 22-year-old harbour seal (Phoca vitulina geronimensis) presented with a subcutaneous mass on the right dorsal carpus. The mass measured 5 cm by 2 cm and was firmly attached to the underlying tissues. Cytology from fine-needle aspiration was consistent with a soft tissue sarcoma. Surgical resection was attempted, however complete excision was prohibited by extensive local invasion. Histopathology results confirmed a moderately cellular, infiltrative, non-encapsulated giant cell sarcoma. Subsequent surgical amputation of the forelimb was performed successfully, with the animal making a full recovery. No radiographic evidence of reoccurrence was identified three years following the surgical procedure. This is the first described case of forelimb amputation in a pinniped. When complete tumour resection is unattainable, forelimb amputation may serve as an alternative or adjunct to medical management of neoplasia in phocids.

The marginal lymph nodes of the dolphin thorax provide lymphatic drainage to the lungs and can indicate pulmonary disease. This study standardized a technique for rapid, efficient, and thorough ultrasonographic evaluation of the marginal lymph nodes in bottlenose dolphins ( Tursiops truncatus). Thoracic ultrasonography was performed on 29 clinically healthy adult bottlenose dolphins. Reference intervals for lymph node dimensions and ultrasonographic characteristics of marginal lymph nodes were determined from four transducer orientations: longitudinal, transverse, oblique, and an orientation optimized to the ultrasonographer's eye. The relationship between lymph node dimensions and dolphin age, sex, length, weight, origin, and management setting (pool versus ocean enclosure) were also evaluated.     Complete ophthalmic examinations were performed on 160 Macaroni penguins ( Eudyptes chrysolophus ) and 90 Rockhopper penguins ( Eudyptes chrysocome ) at eight North American zoological institutions. Cataract prevalence in the Macaroni population was 46.5% (n = 74) of penguins and 42.3% (135/319) of eyes. Cataract prevalence in the Rockhopper population was 45.5% (n = 40) of penguins and 40.6% (73/180) of eyes. The mean age of Macaroni penguins without ocular disease was 7.4 ± 5.8 yr, while that of Rockhoppers was 9.8 ± 6.4 yr. Risk factors for cataract were examined through husbandry surveys completed by each institution and by evaluation of light intensity and ultraviolet (UV) light measurements acquired in each penguin exhibit.     Multiple black raised nodular masses were noted on the dorsal surface of an adult male Haller's round ray Urobatis halleri. Biopsy of 2 masses was performed, and histopathology revealed proliferative sheets of melanocytes exhibiting mild anisocytosis and anisokaryosis, supporting a diagnosis of malignant melanoma. Approximately 2 mo following the biopsy procedure, the round ray became acutely anorexic and was found dead in its enclosure. A full necropsy was performed, and tissues were submitted for histopathology. The black raised nodular masses again exhibited histologic features of a melanoma. In addition to the nodular masses present, multiple flat areas of increased pigmentation were also present throughout the course of the case and were not suggestive of neoplasia histologically. The transformation of benign to malignant neoplasia has been well described in other species and may have played a role in the development of multiple tumors in this case.

    Ophthalmic examinations were performed on 160 macaroni penguins (Eudyptes chrysolophus) and 90 southern rockhopper penguins (Eudyptes chrysocome) at eight North American zoos and aquaria. Intraocular pressure (IOP) was measured using rebound tonometry while penguins were held in two different body positions. Correlations between IOP and factors including age, body position, eye pathology, and housing parameters were evaluated. Normal macaroni penguins had a mean IOP of 42.0 ± 9.7 mm Hg. Normal rockhopper penguins had a mean IOP of 32.9 ± 6.2 mm Hg. Neither species had significantly different IOP between sexes or between left and right eyes of the same penguin. In both species, there was a negative linear correlation between age and IOP.     For wild belugas (Delphinapterus leucas), gestation length estimates based on fetal size have produced extreme ranges. Ex situ populations thereby provide unique opportunities to define this important life history event. Accordingly, research with ultrasound was conducted on six beluga whales over 11 gestations with known conception dates to serially measure fetal changes in biparietal diameter (BP), thoracic diameter (TD), thoracic circumference (TC), and total length (TL). Incremental polynomial regression analyses were performed on each fetal measurement to develop predictive models for determining age based on fetal size and days from parturition. Gestation length (n = 11) was a mean 467 ± 5.4 d with male calves (478 ± 8.6 d) experiencing a longer gestation (P = 0.04) than females (457 ± 3.9 d). Age at TL was best described using a 2nd order polynomial model, while linear relationships existed for BPD, TD, and TC. Accuracy was improved for predicting age (P = 0.001) or days prior to parturition (P = 0.038) using data from the first vs. the second half of gestation. The results provide accurate models for aging beluga fetuses based on size in both in situ and ex situ populations.

    The Omega-3 Index [red blood cell (RBC) content of eicosapentaenoic acid (EPA)+docosahexaenoic acid (DHA)] is inversely related to risk of cardiovascular disease in humans. In the U.S., the average Omega-3 Index is about 4-6% of RBC fatty acids, whereas in Japan it is 9-10%. The range of physiologically-possible levels for the Omega-3 Index in other mammals is unknown. To compare the RBC fatty acid composition of a common piscivorous mammal, the bottlenose dolphin (Tursiops truncatus), with that of (U.S.) humans, and to examine the extent to which dietary fatty acid patterns were reflected in RBCs. RBCs were isolated from routine blood samples collected from 35 healthy dolphins at two display facilities and were analyzed by gas chromatography. For humans, historic, deidentified RBC fatty acid data from our laboratory were used (n=11,329; mean age 58). The mean Omega-3 Index of the dolphins was 19.9% compared with 6.0% for humans. EPA levels were 15.3% vs 1.2%, respectively, but DHA levels were virtually identical (4.6% vs 4.8%). Linoleic acid (LNA) levels were much lower in dolphins vs humans (0.5% vs 12.5%) whereas arachidonic acid (ARA) levels were similar (12.3% vs 14.5%). In a subgroup of humans with an Omega-3 Index in the >99.2 percentile, the mean index was similar to that of the dolphins. Based on an analysis of their food, the dolphins consumed about 60g of EPA+DHA per day as compared to about 0.1g in humans. Dolphins have an Omega-3 Index that is (only) 3-4× higher than that of U.S. adults despite their intake of EPA+DHA being about 165× higher (as a percent of kcal). RBC, EPA and LNA levels are relatively more reflective of dietary intakes than are DHA and ARA levels. The mechanisms by which certain fatty acid levels appear to be fixed and others may vary in RBC membranes are unknown.

    In humans, fetal echocardiography represents the most important tool for the assessment of the cardiovascular well-being of the fetus. However, because of logistic, anatomic, and behavioral challenges, detailed fetal echocardiographic evaluation of marine mammals has not been previously described. Because the application of fetal echocardiography to cetaceans could have both clinical and academic importance, an approach to evaluating the fetal dolphin's cardiovascular status was developed with conventional, fetal echocardiographic techniques developed in humans. Eight singleton fetal bottlenose dolphins (Tursiops truncatus) were evaluated, each between 6 and 11 mo gestation; six fetuses underwent two fetal echocardiographic evaluations each, four at 3-mo intervals, and two at 0.5-mo intervals. Evaluations were performed without sedation, using conventional, portable ultrasound systems. Multiple transducers, probes, and maternal dolphin positions were used to optimize image quality. Fetal echocardiography included two-dimensional imaging and color flow mapping of the heart and great arteries, as well as pulsed Doppler evaluation of the umbilical artery and vein. Thorough evaluations of the fetal dolphins' cardiovascular status were performed, with the greatest resolution between 8 and 9 mo gestation. With the use of published human fetal echocardiographic findings for comparison, fetal echocardiography demonstrated normal structure and function of the heart and great arteries, including the pulmonary veins, inferior vena cava, right and left atria, foramen ovale, tricuspid and mitral valves, right and left ventricles, ventricular septum, pulmonary and aortic valves, main pulmonary artery and ascending aorta, and ductus arteriosus. Pulsed Doppler techniques demonstrated normal umbilical arterial and venous waveforms, and color flow mapping demonstrated absence of significant valvar regurgitation. Fetal echocardiography, particularly between 8 and 9 mo gestation, can provide a safe and detailed assessment of the cardiovascular status of the fetal bottlenose dolphin.
