    The MMPI was administered to 40 undergraduate students with 20 subjects asked to imagine that they were applying for a job and 20 subjects asked to imagine that they were applying for psychotherapy. Subjects in the job group dissimulated (faked good) and those in the therapy group malingered (faked bad) as evidenced by differences between the subtle and obvious scores. The combined subtle and obvious score was higher for the psychotherapy group. It was concluded that the subtle, obvious, and combined subtle and obvious scores on the MMPI were a function of the demand characteristics of the testing situation.

    The results of this study challenge the widely held view that growth hormone (GH) acts only during the postnatal period. RNA phenotyping shows transcripts for the GH receptor and GH-binding protein in mouse preimplantation embryos of all stages from fertilized eggs (day 1) to blastocysts (day 4). An antibody specific to the cytoplasmic region of the GH receptor revealed receptor protein expression, first in two-cell embryos, the stage of activation of the embryonic genome (day 2), and in all subsequent stages. In cleavage-stage embryos this immunoreactivity was localized mainly to the nucleus, but clear evidence of membrane labeling was apparent in blastocysts. GH receptor immunoreactivity was also observed in cumulus cells associated with unfertilized oocytes but not in the unfertilized oocytes. The blastocyst receptor was demonstrated to be functional, exhibiting the classic bell-shaped dose-response curves for GH stimulation of both 3-O-methyl glucose transport and protein synthesis. Maximal stimulation of 40-50% was seen for both responses at less than 1 ng/ml recombinant GH, suggesting a role for maternal GH. However mRNA transcripts for GH were also detected from the morula stage (day 3) by using reverse transcription-PCR, and GH immunoreactivity was seen in blastocysts. These observations raise the possibility of a paracrine/autocrine GH loop regulating embryonic development in its earliest stages.

    An experimental study of steepland channel response to episodic erosion was performed in a large flume. A study in the Little South Fork of the Cache la Poudre River, Larimer County, Colorado, was carried out during the 1979 snowmelt runoff season in order to investigate, under field conditions, phenomena observed in the flume study. Steepland drainage basins can be divided into three zones (1,2,3), on the basis of sediment supply, availability of energy, and the presence or absence of confining valley walls. Zone 1 channels are sites of sediment introduction and temporary storage of coarse sediment, which is eventually mobilized and transported downstream to Zone 2. Zone 2 channels are essentially conduits for sediment transported from Zone 1. Large in-channel fans are formed at channel confluences. The development of a single incised channel on the fans indicates that the sediment stored in Zone 2 is exhausted as does the development of headcuts in the channel themselves. The morphology of Zone 3 channels is dependent on the type and quantity of sediment delivered to them from Zone 1 via Zone 2 channels.

    A three-way crossover study was undertaken in 10 healthy subjects to characterize the reported disulfiram-like activity of moxalactam and to assess its influence on ethanol and acetaldehyde metabolism. On different occasions separated by at least 2 wk subjects were given in random order: 0.5 gm/kg ethanol orally, 0.5 gm/kg ethanol followed in 1 hr by 1.0 gm IV moxalactam, and 1.0 gm IV moxalactam every 8 hr for four doses followed by 0.5 gm/kg ethanol. Mean ethanol elimination rates of 13.1 +/- 0.76, 10.1 +/- 1.11, and 10.9 +/- 1.06 mg/dl/hr (mean +/- SEM) were observed in the three protocols, respectively. Corresponding mean estimated acetaldehyde clearance rates were 103.7 +/- 15.55, 92.8 +/- 13.79, and 97.3 +/- 10.41 l/min (mean +/- SEM). While no consistent moxalactam effect on ethanol or acetaldehyde elimination was observed, two subjects experienced mild disulfiram-like reactions to ethanol after moxalactam pretreatment.     Female Japanese quail received either hexachlorobenzene (HCB, 100 mg/kg . d) or tetrachlorohydroquinone (TCHQ, 175 mg/kg . d) for 10 d as a primary treatment. Following this, a secondary treatment of HCB (100 mg/kg . d) or TCHQ (175 mg/kg . d), alone or in combination, was administered for 1, 5, 10 or 15 d. The primary HCB treatment caused elevated delta-aminolevulinic acid synthetase (ALA-S) activities and small increases in porphyrin concentrations. Subsequent treatment of these birds with lactose resulted in no further increases in porphyrins or ALA-S. TCHQ treatment caused increases in porphyrins similar to those seen with continued HCB treatment. Apparently, despite the fact that TCHQ alone had no affect on ALA-S or porphyrin levels, this compound is able, in the presence of elevated ALA levels to cause porphyria. A combination of HCB and TCHQ administered to HCB-pretreated animals caused a more severe porphyria than did follow-up treatment with either HCB or TCHQ alone.

    The use of non-invasively measured pulse transit time (PTT) to monitor the cardiovascular systems in critically ill patients, like sepsis, can be of significant clinical value. In this study, the potential of PTT and its variability in cardiovascular system monitoring in a mechanically ventilated and anesthetized rabbit model of endotoxic shock was assessed. Eight adult New Zealand white rabbits, which were treated with endotoxin bolus infusion, were studied. Measurements of PTT, pre-ejection period (PEP), and vascular transit time (VTT) were obtained in pre- and post-intervention stages (before and 90 minutes after the administration of endotoxin). The decrease in mean PTT (p < 0.05) and PEP (p < 0.01) in the post-intervention stage reflected sympathetic activation, whilst the increase in respiratory variation in PTT (p < 0.01), PEP (p 〈 0.01), and VTT (p < 0.01) could be attributed to an enhancement of respiratory variation in stroke volume associated with hypovolemia in endotoxic shock. The relationship between beat-to-beat variability in PTT and all other cardiovascular time series were further investigated through linear regression analysis, which revealed that PTT was most strongly correlated with VTT (R(2) ≥ 0.84 with positive slope). Computation of coherence and phase shift in the ventilating frequency band (HF: 0.50 - 0.75 Hz) showed that the respiratory variation in PTT was synchronized with both PEP and VTT (coherence > 0.84 with phase shift less than one cardiac beat). These results highlighted the potential value of PTT and its respiratory variation in characterizing the pathophysioloigcal hemodynamic change in endotoxic shock.

    Several growth factor ligand and receptor gene products have been shown to play roles during preimplantation mammalian development. Genes for insulin-like growth factors (IGFs), transforming growth factors (TGFs), fibroblast growth factor (FGF), platelet-derived growth factor (PDGF) and receptors for insulin, IGF, PDGF, TGF alpha and epidermal growth factor (EGF) are expressed by early embryos of several species including mouse, rat, cow and sheep. Roles of growth factors during early development have been demonstrated by addition of purified growth factors to culture medium or by molecular genetic techniques that interfere with gene expression. In this way, it has been shown that successful development of the blastocyst is dependent on the action of epidermal growth factor (EGF) and leukaemia inhibitory factor (LIF). Recent experiments show that both LIF and EGF stimulate secretion of urokinase-type plasminogen activator (uPA) and gelatinase B/matrix metalloproteinase-9 (MMP-9) in day 7 mouse blastocyst outgrowths. At the same time, tissue inhibitors of MMPs (TIMPs) are also expressed by embryonic, decidual and uterine tissues during the implantation process. It appears that LIF may act directly or indirectly, by inducing the expression of other cytokines, to regulate the temporal and spatial production and activity of proteases and protease inhibitors to create a favourable environment for implantation.

    Phylogeographic inference has typically relied on analyses of data from one or a few genes to provide estimates of demography and population histories. While much has been learned from these studies, all phylogeographic analysis is conditioned on the data, and thus, inferences derived from data that represent a small sample of the genome are unavoidably tenuous. Here, we demonstrate one approach for moving beyond classic phylogeographic research. We use sequence capture probes and Illumina sequencing to generate data from >400 loci in order to infer the phylogeographic history of Salix melanopsis, a riparian willow with a disjunct distribution in coastal and the inland Pacific Northwest. We evaluate a priori phylogeographic hypotheses using coalescent models for parameter estimation, and the results support earlier findings that identified post-Pleistocene dispersal as the cause of the disjunction in S. melanopsis. We also conduct a series of model selection exercises using IMa2, Migrate-n and ∂a∂i. The resulting ranking of models indicates that refugial dynamics were complex, with multiple regions in the inland regions serving as the source for postglacial colonization. Our results demonstrate that new sources of data and new approaches to data analysis can rejuvenate phylogeographic research by allowing for the identification of complex models that enable researchers to both identify and estimate the most relevant parameters for a given system.
