equivalent or better than our current Pacific Blue® reagent and other 450 nm emitting dyes in fluorescence intensity, stability, and overall performance and therefore will replace Pacific Blue® in our product portfolio.

  Attention: Replacement of all PE-Cy5.5 products with PerCP-eFluor® 710 or PerCP-Cy5.5
  The PerCP-eFluor® 710 tandem dye has been developed as an alternative choice for the FL3 channel on a blue (488 nm) laser line. PerCP-eFluor® 710 is consistently brighter than PerCP-Cy5.5 when evaluated in clone to clone comparison tests on identical tissue samples. Although PerCP-eFluor® 710 is slightly red-shifted in its emission compared to PerCP-Cy5.5, it uses the same filter sets as those recommended for PerCP-Cy5.5 detection thereby requiring no changes to instrumentation for customers currently using PerCP-Cy5.5 or PE-Cy5.5 tandem dye conjugates. The PerCP-eFluor® 710 tandem dye is very stable and testing indicates that exposure to ambient light and standard fixation protocols have minimal effects on compensation requirements for this fluorochrome.

The emission spectra of a PerCP-eFluor® 710 (red) conjugated antibody and a PerCP-Cy5.5 (pink) conjugated antibody are overlayed. For both dyes, a 685 nm dichroic mirror is recommended along with a 695/40 or 710/40 band pass filter.

Anti-mouse CD4 (clone RM4-5), anti-mouse CD8 (clone 53-6.7), and anti-human CD3 (clone OKT3) were conjugated to either PerCP-eFluor® 710 (red) or PerCP-Cy5.5 (pink) for direct comparison. Mouse splenocytes were stained with anti-CD4 (left panel) or anti-CD8 (middle panel). Human PBMCs were stained with anti-CD3 (right panel).

  PerCP-eFluorŽ 710 Stability  
PerCP-eFluorŽ 710 Compensation Matrix

PerCP-eFluor® 710 is minimally affected by fixation or exposure to light.

Human PBMCs were stained with anti-CD4 PerCP-eFluor® 710 and analyzed immediately (red). The sample was then divided and either exposed to ambient light for 6 hours on the benchtop (blue) or fixed in 2% paraformaldehyde overnight at 4°C (green) and analyzed again. The compensation and histogram data demonstrate that these treatments have minimal effects on brightness or compensation requirements for the PerCP-eFluor® 710 tandem dye.

APC-eFluor® 780
 
APC-eFluor® 780 tandem dye is a high performance fluorochrome for multicolor flow cytometry. APC-eFluor® 780 is optimally excited by the red laser (633 nm) and has an emission peak of 780 nm. With similar spectral properties to the APC-Alexa Fluor® 750 tandem dye, APC-eFluor® 780 requires no changes to instrument configuration for use.
 
The APC-eFluor® 780 tandem dye can be easily compensated when used with APC and thus offers a quality reagent for use in multicolor staining panels. APC-eFluor® 780 will be replacing APC-Alexa Fluor® 750 in our product portfolio.
