Staining of prelysed normal human peripheral blood cells with Anti-Human IgM Purified followed by Anti-Mouse IgG FITC. Appropriate isotype controls were used (open histogram). Cells in the lymphocyte population were used for analysis.

Description
The SA-DA4 monoclonal antibody reacts with the μ heavy chain of human IgM. It does not react with other classes of human immunoglobulin including IgD, IgG, or IgA. IgM is expressed intracellularly, during early stages of B lymphopoiesis, and then on the surface of mature B cells.
Applications Reported
The SA-DA4 antibody has been reported for use in flow cytometric analysis, immunohistochemical staining of frozen tissue sections, and ELISA. It has also been reported for use in affinity purification of human IgM.
Applications Tested
The SA-DA4 antibody has been tested by flow cytometric analysis of human peripheral blood leukocytes. This can be used at less than or equal to 1 μg per test. A test is defined as the amount (μg)/test of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 105 to 108 cells/test. It is recommended that the antibody be carefully titrated for optimal performance in the assay of interest.
