Description
The R22/7 monoclonal antibody reacts with the amino terminus of human CCR5 (CD195), an approximately 45 kDa member of the G protein-coupled receptor (GPCR) superfamily that binds chemokines including MIP-1a, MIP-1ß and RANTES. Human CCR5 is expressed on resting T cells with a memory/effector phenotype, monocytes, macrophages and immature dendritic cells. CCR5 functions as a receptor to direct the chemotaxis of lymphocytes, and is the primary co-receptor for macrophage-tropic Human Immunodeficiency Virus Type 1 (HIV-1), which binds CCR5 through gp120.

Applications Tested
This R22/7 antibody has been pre-titrated and tested by flow cytometric analysis of human peripheral blood leukocytes. Refer to catalog number suffix on the vial for amount to use per test: 71/73 are 20 μL (0.125 μg) per test; whereas 41/42 are 5 μL (0.125 μg) per test. A test is defined as the amount (μg) of antibody that will stain a cell sample in a final volume of 100 μL. Cell number should be determined empirically but can range from 105 to 108 cells/test.
Special Notes
R22/7 stains with lower intensity on human peripheral blood mononuclear cells, than on human lysed whole blood. For staining human peripheral blood mononuclear cells, we recommend the use of clone eBioT21/8 (cat. 1957).
